Blunt/TA Ligation Master Mix
The Blunt/TA Ligation Master Mix is developed for ligation of difficult DNA fragments as a ready-to-use 2X master mix containing T4 DNA Ligase and ligation enhancers. The proprietary components of the master mix significantly improve ligation of difficult DNA fragments with either blunt-ends or single-base overhang ends.

The Blunt/TA Ligation Master Mix makes it easy to set up ligation reactions by simply mixing the DNA fragments with the 2X Master Mix. The ligation of all types of DNA ends is fast at room temperature with high efficiency. The subsequent transformation protocol is also simplified to save both time and efforts.
Ligations with single-base overhangs using the Blunt/TA Ligation Master Mix with reactions for different times at 20°C (left) or at different temperatures for 15 minutes (right). Transformants resulting from triplicate plating indicate that the Master Mix works well at room temperature (20°C-25°C) in 15 minutes.
Comparison of T4 DNA ligase (high concentration), the Rapid Ligation Master Mix (Cat.# 40102) and Blunt/TA Ligation Master Mix (Cat.# 40101) for ligation of nick, sticky end, 3´ single-base overhang, and 5´ single-base overhang substrates. All ligations were incubated for 15 minutes except for the 5´ single-base overhang, which was incubated for 1 hour.
